ABSTRACT |
The present study was made with the aims of establishing an acceptable nasal allergy model and presenting the basic data for further studies. Nineteen guinea pigs(alergy group) were sensitized intraperitoneally using 20μg of ovalbumin as an allergen and 10mg of Al(OH)3 gel as an adjuvant in 1ml of normal saline 5 times at intervals of 2 weeks. After then, intranasal sensitization was performed by 1 minute-ultranebulization of 0.1% ovalbumin solution 5 times per week for 4 weeks. One day later, nasal provocation was performed by nasal instillation with 50μl of 1% ovalbumin solution. As control group, 16 guinea pogs were used. The titers of homocytotropic antibodies were identified by passive cutaneous anaphylaxis (PCA) test and the symptoms and signs, the peripheral blood, and the histophatology of the nasal mucosa from the inferior conchae were examined in both groups. The results were as follows ; 1) The titers of three homocytotropic antibodies(IgGla, IgG1b, IgE) by PCA test were all significantly higher in allergy group than in control group. 2) The number of eosinophils, goblet cells, and mast cells in the nasal mucosa were significantly larger in allergy group than in control group. 3) The epithelial loss of nasal mucosa in allergy group was significantly more than that of control group. 4) There were no significant differences in the symptom scores for 30 minutes and in the number of peripheral blood eosinophils between two groups. Based on the results, it was concluded that an animal model of nasal allergy was developed by identifying the presence and the titer of the homocytotropic antibodies and obverving many findings compatible to nasal allergy described above. It was quite meaningful to be able to get allergic conditions with simultaneity and relative homogeneity from the allergy models.
